BTI Cell Growth Factors & Attachment Factors BTI has been manufacturing growth factor (GF) and attachment factor (AF) products since 1981. Our extensive product line includes receptor-grade, aseptically packaged GFs and AFs. These are highly purified proteins that have been membrane filtered, aseptically filled and lyophilized. Most BTI GFs have been tested in cell culture, receptor binding studies and in immunoassays. Some GFs are available [I125] labelled. anti-Human EGF, Rat EGF EGF from human and mouse sources is significantly different from rat EGF. Antibodies directed to human or mouse EGF do not recognize rat EGF sufficiently to adequately assay, localize or inhibit rat EGF containing samples. anti-Rat EGF, ASSOCIATED PRODUCTS: Endothelial Mitogen (ECGS) Endothelial Mitogen (ECGS) is a partially purified preparation from bovine hypothalamus. Endothelial Mitogen was originally prepared to facilitate the growth and survival of human (or mammalian) vascular endothelial cells in vitro. BTI ECGS is now used to improve growth, supplement and/or reduce serum concentrations in cultures of endothelial cells, Balb/C-3T3 cells and in hybridoma cell cloning. Optimal concentration range is from 25-200ug/ml and BTI ECGS does not contain streptomycin which may be toxic to cells. Acidic FGF Acidic FGF, a member of the Class l heparin-binding growth factors (Lobb et al, JBC, 261, 1924, 1986) is structurally and functionally nearly identical to endothelial cell growth factor (ECGF), eye derived growth factor-II, and RDGF. The peptide has potent angiogenic activity and is mitogenic, potentiated by heparin, for vascular endothelial cells and other mesoderm-derived cells. BTI offers both bovine acidic FGF and human recombinant acidic FGF. BTI Bovine aFGF is purified to near homogeneity from bovine hypothalami using heparin-sepharose affinity chromatography (Burgess et al, JBC, 260: 11389, 1985). The BTI human aFGF (15.5kDa) is produced in E.Coli (1-140 a.a.) and purified to near homogeneity. Both products are membrane filtered and aseptically lyophilized. Acidic FGF, Bovine Acidic FGF, Human Basic FGF Basic FGF, a Class II heparin-binding growth factor is characterized by an isoelectric point 9.6 and elution from heparin-sepharose at 1.6-1.8M NaCl. bFGF has similar biological activities to aFGF, but mitogenicity is not potentiated by heparin. BTI prepares both bovine and human bFGF. BTI Bovine bFGF is purified (~95%, SDS-PAGE) from bovine pituitary glands via heparin sepharose and ion exchange chromatography. Our human bFGF (m.wt. 17,304) is produced in E.Coli and purified to near homogeneity. Both products are membrane filtered and aseptically lyophilized. Basic FGF, Bovine Basic FGF, Human anti-Basic FGF IgG BTI anti-bFGF is prepared in rabbits from human recombinant bFGF. The IgG is purified from the rabbit immune serum. It recognizes bFGF and not aFGF on Western immunoblots. Greater than 90% inhibition of bFGF (10ng/ml) was demonstrated on BAEC using this antibody at 1:50 to 1:100. bFGF has been localized in a variety of cells and tissues from human, bovine, rat and mouse sources using BTI anti-bFGF IgG. Human IGF-I & II BTI Human IGF-I and Human IGF-II are recombinant DNA produced. Both have full biological activity and are suitable for cell culture or receptor studies. Human IGF-I Human IGF-II Nerve Growth Factor Nerve Growth Factor, 2.5S NGF, is isolated from mouse submaxillary glands. It is purified by a modification of the method of Bocchini and Angeletti (PNAS 64:787, 1969) to greater than 95% (PAGE). Our NGF is bioassayed in a rat pheochromocytoma cell line. 2.5S NGF is membrane filtered and packaged as a lyophilized solid. Catalog No: BT-206 anti-NGF BTI anti-NGF is produced in rabbits from our mouse 2.5S NGF. The resulting antibody is purified by absorption. 90% inhibition of NGF (10ng/ml) activity was demonstrated on PC-12 cells at 1:100 dilution. Western blots of mouse 2.5S NGF (5ng) are detectable at 1:200 dilution of our anti-NGF using GAR alkaline phosphatase detection. BPE also known as Keratinocyte Growth Supplement is used in conjunction with other growth factors, hormones and media to reduce or eliminate serum in the culturing of some epithelial cell types. BPE has been effectively utilized in culturing normal human epidermal keratinocytes (Tsao, M.C., et al, J. Cell Physiol. 110: 219-229, 1982) and other epithelial cells. Human Vascular Endothelial Cell Growth Factor, h-VEGF, is an angiogenesis factor produced by vascularized tissue. surface glycoprotein and is also present in blood plasma. Fibronectins
are used to facilitate the attachment of cells to a variety of surfaces
and to maintain cells in low serum conditions. Fibronectin from three
different species are purified from plasma to 95% (5% SDS PAGE) and provided

can effectively measure levels of human fibronectin to 50 nanograms per

Many lyophilized fibronectin preparations are difficult to solubilize (recoveries are only 85-95% at best) and after solubilization these preparations tend to aggregate and precipitate on standing. The BTI Fibronectin Solutions are completely solubilized and are stabilized to prevent precipitation. They retain full biological activity and are stable for 9 months stored at 4oC.
