Cyclic AMP & Cyclic GMP EIA Kits

About The Assays

The BTI Cyclic AMP and Cyclic GMP EIA Kits are complete systems.  Each kit uses alkaline phosphatase labeled tracer (cAMP or cGMP) competing for sites on the antibody with standard or sample.  Both assays employ a highly specific antibody developed by BTI to insure accurate, consistent results.  The assay sensitivities are outstanding.  As little as 0.005 picomoles per well of cAMP or cGMP can be measured.

Both kits include all the necessary reagents (plus two coated 96-well strip ELISA plates) to perform the tests either acetylated or unacetylated.  Levels of cAMP or cGMP can be conveniently measured from most biological samples (urine, plasma and cell culture) without the extensive purification procedures inherent in other methodologies.

These are the most versatile cAMP and cGMP kits available. Different sample types can be accommodated easily ( ie. Cell culture supernates and cell extracts(~p[H] 7) ) without sacrificing specificity or sensitivity. BTI scientists have extensive experience in working with a large variety of samples and are available for consultation.

Both the Cyclic AMP and Cyclic GMP assays can be run either overnight or in 6 hours.  The protocol includes only two pipeting steps and one wash sequence per assay.  Each kit accommodates 80 samples run in duplicate (or 160 wells for unknowns).

The shelf life of these BTI Cyclic EIA Kits is 5 months stored at 4ºC.

Procedural Outline
(for both Cyclic AMP and Cyclic GMP EIA Kits)

1. Add samples/standards, tracer and antibody to appropriate wells.  Cover and
    incubate at 4ºC overnight or at 20ºC for 4 hours.

2. Aspirate and wash five times.

3. Blot excess liquid.  Add 200ul substrate to all wells, cover, incubate for 2 hours at 37ºC.
    Measure absorbance at 410nm.

Human EGF EIA Kit

About The Test

The BTI Human EGF EIA Kit employs a competitive binding technique in which biotinylated hEGF competes with unlabelled hEGF for a limited number of specific binding sites on the immobilized antibody.  The protocol is 3.5 hours and requires only a 50ul sample.  The assay range is 0.5-100ng/ml.  Urine samples can be assayed directly without dilution.  All the necessary reagents, a human urine control, and a complete protocol are included with the kit.

Human Fibronectin Elisa Kit

Introduction

Fibronectin, a major cell surface glycoprotein, is found on cell surfaces and circulating in plasma.  It has been shown to be important in wound healing and in cellular interactions.

The BTI Human Fibronectin ELISA Kit measures human fibronectin from both plasma and cellular extracts.  The assay employs a standard 3.5 hour Elisa protocol.  A 100 microliter sample is required and the range is 25-2000 nanograms of human fibronectin per ml.

A number of other species of fibronectin have been evaluated with the kit.  The range of these other species' assays is less sensitive.  Bovine Fibronectin is 0.5-100ug/ml, Mouse Fibronectin is 0.2-50ug/ml, and Rat Fibronectin is 0.2-20ug/ml.

1. Add 100ul samples or standard in duplicate, add 50ul of Fibronectin Ab. Incubate at 37ºC.

2. Add 50ul alkaline phosphatase HFN conjugate. Incubate at 37ºC for 1 hour.

3. Aspirate and wash (3x).

4. Add 200ul Substrate, incubate @37deg.C 1hour. Read Absorbance at 410nm.

Human Involucrin Elisa Assay

About The Assay

Involucrin, a soluble cytoplasmic protein with a highly repetitive amino acid sequence, serves as a substrate for transglutaminase cross-linking in the formation of cornified envelopes.  Like the cross-linked envelope, involucrin expression is a specific marker for keratinocyte differentiation.  This protein is synthesized not only by cells of the epidermis and other squamous epithelia, but also in epithelia undergoing squamous metaplasia and in certain tumors, as in the lung, which display epidermoid characteristics.  In culture the growth and differentiation of keratinocytes, especially those derived from tumors, can be modulated greatly by physiological (retinoids, calcium) and toxic agents (including carcinogens).  The involucrin content of the cells is highly responsive to such treatments and thus its measurement by ELISA can serve as a convenient and sensitive indicator of the cellular response.

The BTI Involucrin ELISA Kit is designed to provide a reproducible, quantitative method of measuring involucrin. The assay is highly sensitive, 0.05-0.5  nanograms per well.  Levels of human involucrin can be directly measured after separation of cellular debris.  The kit includes highly purified Human Involucrin, anti-Human Involucrin (rabbit) and Goat anti-Rabbit IgG Alkaline Phosphatase. A complete protocol is provided with each kit.

Procedural Outline

1. Simultaneously incubate (4ºC) overnight Involucrin in Plate No.1, and Antibody + Standards or Antibody + Unknowns
    in Plate No.2.

2. Transfer contents of Plate No.2 to Plate No.1. Incubate at room temperature 30 minutes.

3. Rinse Plate No.1, add GAR IgG Alkaline Phosphatase, incubate 1 hour at room temperature.  Rinse and
    add developing agent, incubate 1-4 hours.

4. Read Plate at 405nm.

HUMAN LYSOZYME EIA KIT

About The Assay

Lysozyme (muramidase) is an enzyme present in serum, plasma, amniotic fluid, milk, saliva, tears, urine and many other biological fluids.  Lysozyme breaks down the walls of certain bacteria and protects against bacterial infections.  Lysozyme has recently been reported as having anti-viral activity against HIV type 1.  Elevated lysozyme levels in urine and serum have been reported in many human disease states, including leukemias (FAB-M4, CMML, CML), tuberculosis, megaloblastic anemias, acute bacterial infections, ulcerative colitis, severe renal insufficiency, pyelonephritis, nephrosis and renal transplant rejection.

The BTI Human Lysozyme EIA Kit is a sandwich ELISA assay.  The assay employs two highly specific lysozyme antibodies.  Our MAb is immobilized in the wells and a second polyclonal antibody is used as the reporter antibody.  The assay is sensitive (0.78-50ng/ml) and specific for human lysozyme.  Each Kit includes all the necessary reagents, precoated 96-well strip plate, urine control and a complete 4 ¼ hour protocol.

Procedural Outline

1. Add 100ul standard/sample/control/blank, cover and incubate at room temperature for 2 hours.

2. Aspirate and wash three times.

3. Add 100ul of lysozyme antibody to each well, and incubate at room temperature for 1 hour.

4. Repeat Step 2, then add 100ul of DAG-HRP to each well, cover, and incubate at room temperature for 1 hour.

5. Repeat Step 2, then add 100ul of substrate to all wells, and incubate for 15 minutes in the dark at room temperature.

6. Add 100ul of stop solution, swirl and read at 450nm.

Intact Human Osteocalcin EIA Kit

About The Assay

Osteocalcin (5800 daltons) is a specific product of the osteoblast.  It is primarily deposited into the ECM of bone and only a small amount of newly synthesized osteocalcin is released directly into the circulation.  Studies suggest that there are various forms of osteocalcin in the circulation and that different antibodies detect both intact and fragments of osteocalcin.  The physiological significance of osteocalcin fragments is unclear but they may be derived from resorption, osteoblastic synthesis, systemic catabolism or all of these.  Since serum proteases may also degrade the intact osteocalcin, it is recommended that blood samples be processed quickly in the presence of protease inhibitors.

The BTI Intact Osteocalcin EIA measures only intact osteocalcin which is synthesized de novo by the osteoblast and it eliminates any interference by circulating fragments.  The assay is a sandwich EIA which employs two monoclonal antibodies directed towards the amino-and the carboxy-terminal regions of the human osteocalcin.  It recognizes only intact osteocalcin, requiring the full 49 residue protein for detection. This assay reacts with human, bovine, monkey and dog osteocalcin.

The assay is highly sensitive (Range: 1-50ng/ml)  and requires only a 25ul sample.  All the necessary reagents, a 96-well strip plate, and a complete 3½ hour protocol are included with the kit.

Procedural Outline

1. Add 25ul standard/sample.  Add antibody and gently swirl.  Incubate 2½ hours at 37oC.

2. Aspirate and wash 3 times with wash buffer.

3. Add Strep-Av-HRP, swirl, incubate ½ hour at room temperature.

4. Repeat Step 2. Then add 100ul of substrate, swirl, develop 10 minutes in the dark.
    Add 100ul stop solution, swirl, measure absorbance at  450nm.

Mid-Tact Human Osteocalcin EIA Kit

About The Assay

Osteocalcin (5800 daltons) is a specific product of the osteoblast.  Only a small amount of osteocalcin is released directly into the circulation, it is primarily deposited into the bone matrix.  Studies have shown that osteocalcin circulates both as the intact (1-49) protein and as N-terminal fragments.  The major N-terminal fragment is the peptide (1-43).

The BTI Mid-Tact Osteocalcin Elisa Kit is highly specific. It measures both the intact human osteocalcin and the major (1-43) fragment.  The assay is a sandwich EIA which employs two monoclonal antibodies.  One antibody (1-19) is immobilized in the wells and the second antibody (30-40) is biotinylated.  The assay is highly sensitive (0.5ng/ml) and requires only a 25 microliter sample.  All the necessary reagents, a 96-well strip plate, and a complete 3½ hour protocol are included with the kit.

BTI Rat Osteocalcin EIA Kit


About the Test

The BTI Rat Osteocalcin EIA Kit is a sandwich ELISA assay.  Two osteocalcin antibodies are employed, each directed toward an end of the (C or N terminal) osteocalcin molecule.  The N-terminal monoclonal antibody is bound to the wells and when reacted with rat osteocalcin standard or sample, the C-terminal rat osteocalcin antibody completes the sandwich.  This ELISA kit shows good  correlation (r = 0.95) with our rat osteocalcin RIA.

This sandwich ELISA kit is specific for rat osteocalcin only.  Both carboxylated and decarboxylated rat osteocalcin are recognized. Rat osteocalcin can be measured directly from serum, heparinized plasma, bone extracts and serum-free cell culture supernates.

The assay can be run either overnight (0.33-20ng/ml) or in 6 hours (0.78-50ng/ml).  As little as 10 microliters of rat serum is required for duplicate determinations.  Each kit contains all the necessary reagents, an authentic rat serum control, precoated 96-well strips and a complete protocol.

Human Osteocalcin RIA Kit

About The Assay

Osteocalcin, the major non-collagenous protein of bone, is a specific product of the osteoblast and its synthesis is stimulated by 1, 25-Dihydroxyvitamin D3. A fraction of that synthesized does not accumulate in bone but is secreted directly into the circulation.  Animal studies indicate serum osteocalcin is derived from new cellular synthesis rather than from the release of bone matrix protein during resorption. Furthermore, serum osteocalcin concentrations are related to histomorphometric measurements of bone formation but not resorption.  Deviations from normal concentrations of circulating osteocalcin are a consequence of changes in the synthesis or degradation of the protein.  Such changes may result from physiological alterations in skeletal homeostasis that accompany normal development or may be associated with specific disease states.  While the precise biological function of osteocalcin is not known, it is the most specific marker of osteoblastic function described
to date (1).

The availability of commercial immunoassay kits and reagents has resulted in a large body of literature regarding serum osteocalcin measurements in normal individuals and in those with a variety of metabolic bone diseases. Unfortunately, the absolute values obtained with these kits deviate substantially (2) from results obtained in various research laboratories.
The BTI Human Osteocalcin RIA Kit offers the advantages of the use of all human reagents and values that are consistent
with those obtained in research laboratories.

The BTI Human Osteocalcin RIA Kit is user friendly.  The kit includes 5 individual standards and a simplified 3.5 hour protocol.  The assay requires only a 50ul sample and yields a range of 1.0-50 nanograms per ml.  Serum samples and cell culture supernatants can be measured directly.

About The Assay:

The BTI Rat Osteocalcin Radioimmunoassay Reagents are designed  to provide an inexpensive, flexible method for the accurate measurement of rat osteocalcin.  The assay is highly sensitive 0.025-1.5 nanograms per tube. Human TGF-alpha RIA Kit

About The Assay

Transforming Growth Factor-alpha (TGF-alpha) is a peptide structurally related to epidermal growth factor (EGF) and also binds the EGF receptor. Synthesis of  TGF-alpha has been reported in several transformed cells and tissues during early fetal development and recently in normal adult keratinocytes.  Addition of EGF or TGF-alpha induces expression of  TGF-alpha messenger RNA, suggesting the presence of an auto-inductive mechanism in the normal regulation of the epidermis. Expression of TGF-alpha could also potentially occur during wound healing and in various pathological states besides neoplasia.

The BTI Human TGF-alpha RIA Kit employs a sensitive, highly specific methodology to measure hTGF-alpha.  The method utilizes a competitive binding technique in which [I125] TGF-alpha compete for binding sites on our highly specific hTGF-alpha antibody.  The simple procedure involves one overnight incubation and yields a sensitivity of 0.02 nanograms.

Included in the kit are all the necessary reagents and a detailed protocol.

About The Test

The BTI Cyclic AMP and Cyclic GMP RIA Kits employ state-of-the-art reagents and methodologies. Both kits have highly specific primary antibodies preconjugated with a second antibody to minimize non-specific binding and to eliminate a second incubation step. The highly immunoreactive Tracers, [I125]S-Cyclic AMP-TME and the [I125]S-Cyclic GMP-TME are purified via HPLC and are monoiodinated.

The assay sensitivities are outstanding. The Cyclic AMP RIA Kit is 0.1 picomoles per tube and 0.005 picomoles per tube for acetylated samples. The Cyclic GMP RIA Kit is 0.2 picomoles per tube and 0.005 picomoles per tube for acetylated samples. Both kits employ a fast, simple protocol.

The protocol includes only three pipetting steps, one incubation, and generates a visible precipitate. All necessary reagents including  true non-specific binding reagents and acetylation reagents are included in each 200 tube kit. Both kits have a shelf life of 8 weeks refrigerated at 4oC.

Levels of Cyclic AMP and Cyclic GMP can be conveniently measured from most biological samples (urine, plasma, tissue and cell culture) without the usual extensive extraction and purification  procedures inherent in other methodologies.

These are the most versatile cAMP and cGMP kits available. Different sample types can be accommodated easily ( ie. 1. (At room temperature) Add buffer, Standard or Sample, Tracer and Antibody to appropriate tubes.
    Mix tubes gently in rack.

2. Incubate at 4oC overnight.

3. Add 1 ml of cold Buffer and centrifuge.

4. Decant or aspirate supernatants. Count each tube.

About The Assay

Osteocalcin, a major non-collagenous bone protein, is synthesized by osteoblasts during bone formation. Most of the osteocalcin is incorporated into the bone matrix and a small amount is secreted into the circulation. Studies have shown that serum osteocalcin levels are directly related to osteoblastic activity and bone formation.

The BTI Mouse Osteocalcin EIA kit is a sandwich ELISA assay. Two mouse osteocalcin antibodies are employed, each directed toward an end (C or N terminal) of the osteocalcin molecule. One mouse osteocalcin antibody is bound to the wells which binds the mouse osteocalcin standard or sample, the other biotinylated antibody completes the sandwich.

This sandwich ELISA kit is specific for intact mouse osteocalcin only. Both carboxylated and decarboxylated mouse osteocalcin are recognized. Mouse Osteocalcin can be measured at a 1:10 to 1:20 dilution from serum or heparinized plasma. Bone extracts and serum-free cell culture supernates can be measured directly. This kit shows good correlation (r=0.90) with the BTI mouse osteocalcin RIA.

The protocol has an overnight incubation and yields a
1-50ng/ml range. As little as 5-10 microliters of mouse serum is required for duplicate determinations. Each kit contains all the necessary reagents, precoated 96-well strips and a complete protocol.

Procedural Outline

1. Add 25ul standard/sample/control/blank to individual wells, then add 100ul of biotin-MOC Ab, cover, incubate at 4ºC for 18-24 hours.

2. Aspirate and wash three times.

3. Add 100ul of HRP-Strep A to each well, cover, and incubate in the dark at rm. temp for 30 minutes.

4. Repeat Step 2, then add 100ul of TMB-Perox. to each well, cover, and incubate in the dark at room temperature for 15 minutes.

5. Add 100ul of stop solution, swirl and read at 450nm.
